Regulation mechanism of soybean glycinin gene expression

Abstract
To study the regulatory mechanism of the plant gene expression. the genomic DNA clone for the soybean glycinin gene was isolated. A soybean genomic library was constructed into EMBL 3 vector and plaque hybridization was carried out with the cDNA clone of glycinin as a probe. The clone lambda G47 contains two glycinin genes, Gy1 and Gy2, in the same orientation. The structure of glycinin gene Gy2 was determined by Southern blot hybridization and nucleotide sequencing. There are 2863 by of coding region encoding 485 amino acids. which consist of 4 exons and 3 introns. The 5¢¥ noncoding region includes consensus sequences considered to be regulatory elements for development and tissue specific expression of the glycinin gene. To identify trans-acting factors and cis-acting elements required for regulation of glycinin gene expression. DNA binding study was carried out with the extract prepared from developing soybean seeds. Molecular nature of the proteins which binds to 5¢¥ noncoding region of the glycinin gene will be discussed.
